Most memorable mission for me has to be The Shalebridge Cradle in Thief 3. That mission just redefined scary for me, I've never felt a level with so much atmosphere and suspense..

Well, there is that one quest in Fallout 2 where you blow up a toilet and cover the whole town in poop.
That was a fun mission.

The first video game quest that comes to mind was the Lungfishopolis level from Psychonauts. The whole idea of playing a Godzilla-esque monster for that level was definitely unique and fun.

RPG quests, especially in a fantasy setting, have a habit of blurring together, but I found it quite cool how so many quests in Planescape: Torment involved ripping off your own body parts.

...then there was the time my friend Zeinab got pissed off because of the quest in Morrowind to get "Zeinab's Thong". She thought I'd modded the game to make a joke on her.
